Abstract
We have described the partial characterization of Protein Kinase-C (PKC) from human uterus with hyperplastic endometrium. The enzyme was eluted at 0.25 M NaCl concentration from DEAE-Sepharose column. PKC was partially dependent on Ca++ and phospholipids. 20 μM tamoxifen has strongly inhibited (about 80%) the enzyme. The Km for ATP was about 23 μM while histones IIS and IIIS showed substrate inhibition above the concentration of 1 mg/ml. Maximum activity was observed in the presence of 10 mM Mg++ in the reaction mixture. ADP as a product showed strong inhibition at 1-3mM concentrations. All phorbol esters and fatty acids tested showed little inhibitory effect while phospholipids (PLs) showed slight activation. This study indicates that hyperplasia of endometrium may be due to altered characteristic of PKC.
